Synergistic antitumour effect of a combination of gene therapy with IR treatment. (A) Ad/EFp-BCD or Ad/5HREp-BCD was intratumorally injected into tumor-bearing mice on day 0 (blue arrow head), and 5-FC was administered daily from day 1 to day 5 (red arrow head). Body weight of the tumour-bearing mice was measured during and after the treatment. The results are the mean of six independent mice±s.d. *P<0.05. **P<0.01. (B and C) Tumour-bearing mice in the Ad & 5-FC group and Ad & 5-FC & IR group were administered Ad/5HREp-BCD. Those in the Ad & 5-FC group, IR & 5-FC group, and Ad & 5-FC & IR group were administered 5-FC. The tumour xenografts in the IR group, IR & 5-FC group, and Ad & 5-FC & IR group were locally exposed to irradiation with a single dose of 12.5 Gy (B) or daily dose of 3 Gy for 5 days (C). (See Materials and methods for details.) Tumour volume was measured with calipers and calculated as 0.5 L W2. Relative tumour volume indicates the ratio of the tumour volume on each day to the corresponding volume on day 0. The results are the mean of six independent tumours±s.d.
