HIF and the VHL complex. Under normoxic conditions, HIF is hydroxylated by HPH. Hypoxia inducible factor 1α prolyl hydroxylases activity requires molecular oxygen, 2-OG, ascorbate (Asc), and iron (Fe). Following hydroxylation (represented by the OH group), HIF is recognised by the VHL complex and targeted for degradation. Pseudohypoxia results from VHL alterations that result in HIF accumulation with upregulation of potentially tumorigenic genes.
