Effect of G-quadruplex interactive agents on VEGF mRNA synthesis in various human cancer cells, including human breast cancer cell line MDA-MB231 (A), human endometrial cancer cell line HEC-A1 (B), and human renal cancer cell lines A498 and 786-O (C, left to right). Human cancer cells were treated with TMPyP2, TMPyP4, and Se2SAP at various concentrations for indicated times, and VEGF mRNA and beta-actin mRNA were measured by RT-PCR. Less than 30% reduction in the number of cells occurred over 3 days after treatment with 50 μM TMPyP4, TMPyP2 and Se2SAP (data not shown). Two products were obtained after RT-PCR, which correspond to c-DNAs of VEGF121, and VEGF165.
