Phospholipase C pathway. P2Y₂-receptor mediated [Ca²⁺]_i-elevations in Kyse-140 cells are mediated by phospholipase C activation. Pre-treatment (10 min) of Kyse-140 cells with the phospholipase C inhibitor U73122 (10–50 μM) dose-dependently inhibited [Ca²⁺]_i elevation in response to ATP (100 μM), whereas the inactive U73122-analogue U73343 (50 μM) did not affect P2Y₂-receptor mediated [Ca²⁺]_i signalling. [Ca²⁺]_i is given as the fluorescence ratio (ΔF340/F380 nm) of the Ca²⁺-sensitive dye fura-2. Representative single-cell tracings out of five independent experiments with 20 or more cells per incubation are shown.
