Figure 6. Cx43−/−-fibs exhibit significantly reduced intercellular coupling.
Fibroblast monolayers loaded with the gap junction permeant dye calcein/AM before (A) and after (B) photobleaching of a single cell within the monolayer. C, the time-dependent fluorescence recovery after photobleaching (FRAP) was recorded for 30 and 50 min, respectively, in WT- (♦; n = 14) and Cx43−/−-fibs (;▪ n = 10). The time constant of recovery was significantly lower in WT-fibs (τ = 246 ± 14 s) in comparison to Cx43−/−-fibs (t = 1140 ± 50 s), an indicator for the significantly reduced intercellular coupling in Cx43−/−-fibs. D, FRAP can be reversibly inhibited by incubation of the Cx43−/−-fibs monolayers with the gap junction inhibitor carbenoxolone (100 μm).
