(A) Glycine uptake was performed as described in Material and Methods with oocytes expressing GlyT1b, GlyT1b-EL1, GlyT1b–C62A, GlyT2a, GlyT2a-EL1 or GlyT2a-A223C. Oocytes were pre-treaded 10 min in cholineCl Ringer with or without MTSET (2 mM). Oocytes were incubated for 10 minutes in 5 ml control NaCl Ringer or cholineCl Ringer solution containing 5 μM U-14C glycine. (B) Charge coupling was estimated as the ratio between the timeintegral of the current evoked by the application (60 s to 90 s) of 25 μM U-14C glycine with 75 μM cold glycine and the simultaneous glycine uptake (see methods).
