(A) Northern blot analysis of the effect of met22-Δ, trf4-Δ, and rrp6-Δ mutations on tRNA levels in trm44-Δ tan1-Δ mutants. Strains were grown in YP glycerol media at 30°C and shifted to 36.5°C for the indicated times. RNA (1.5 μg) was analyzed by Northern blotting. (B) Quantification of the levels of tRNA species 8 h after temperature shift. For tRNASer(CGA), tRNASer(UGA), and tRNASer(IGA), the bars indicate the ratio of tRNA levels after 8 h at 36.5°C, relative to levels in the wild-type strain before temperature shift (each value itself first normalized to 5S RNA).
