Plus-strand transfer and elongation. (A) When the (+) strand intermediate is synthesized (82 nt long), replication terminates. A recipient strand transfer template (60 nt long) added to the reaction anneals to the 3′ end of the (+) strand intermediate and serves as a template for further synthesis to generate a 124-nt product. (B) For strand transfer and elongation reactions, a 60-nt recipient strand transfer template was added along with the initial templates a, b, or c in lanes 5–7, respectively. Reactions lacking the recipient template are shown for comparison in lanes 1–3. The reaction mixtures were incubated for 15 min at 37°C, subjected to electrophoresis, and exposed to x-ray film.
