Transient transfection of endothelial cells with a small interfering RNA (siRNA) for RAGE or the control oligo was carried out using the Amaxa Nucleofection Device (see the Materials and Methods section). Transfection efficiency was measured by the percentage of cells expressing green fluorescence protein (GFP) after transfection with pmaxGFP.
