(A) Wild type M. tuberculosis H37Rv TDM (wtTDM) dampens IL-12 production induced by ΔmmaA4 mutant TDM. Bone marrow-derived macrophages were incubated with ΔmmaA4 TDM alone or with a mixture of purified wild type TDM and ΔmmaA4 TDM. IL-12p40 and TNF-α were measured in culture supernatants by ELISA. *, p<0.05 (one-way ANOVA, Bonferroni post-tests). Values are the means±SD of triplicate samples and are representative of two separate experiments performed on at least two independent batches of purified TDM. (B) M. tuberculosis TDM (wtTDM) dampens IL-12 production induced by LPS. IL-12 p40 and TNF-α accumulation was measured by ELISA in supernatants of bone-marrow derived macrophages stimulated with LPS alone, or with LPS combined with either wild type TDM or ΔmmaA4 TDM. *, p<0.05; **, p<0.01 (one-way ANOVA, Bonferroni post-tests).
