A survey of immunoreactivity for each protein marker was performed after the indicated treatment conditions leading up to, during, and following differentiation with NDM (black bars) or N2/VPA exposure (white bars) at high (40x objective) and low (10x objective) magnification. Scores between 1 and 4 were assigned by investigators blinded to the treatment based on fluorescence intensity and fraction of total cells expressing that particular marker according to the following scale: 4 = intense immunoreactivity in many (> 50 %) of the cells, 3 = strong staining in approximately 50% of the cells, 2 = detectable staining in less than 50% of the cells, 1 = detectable staining in a small fraction (< 10%) of the cells, 0 = no detectable immunoreactivity (Panels B, C, D). Independent of which differentiation medium was used (NDM (black bars) or N2/VPA (white bars), See Materials and Methods), results were similar for each of the different markers.
