Activity of MbCsk-treated MbSrc1. YOP-treated Hck or MbSrc1 (0.8 μm) were incubated in MbCsk assay buffer alone or in the presence of GST-Csk or GST-MbCsk (2 μm) for 15 min at 30 °C. The enzymatic activities of Hck and MbSrc1 were then measured using [γ-32P]ATP and RCM-lysozyme (0.4 mg/ml). Reactions proceeded for 1 min at 30 °C and were analyzed by spotting on Whatman No. 3MM paper, washing in warm 5% trichloroacetic acid, and scintillation counting.
