Higher-dose CCI-779 treatment reversed Pten-deficient hypertrophy in cerebellum. (a) Western blotting of extracts from adult cerebellum from control (+/+) and Pten mutant (–/–) mice treated with vehicle (–), 25, or 50 mg/kg CCI-779 for 4 weeks. At these dosages, CCI-779 treatment effectively decreased downstream phosphorylation of S6. (b) The diameter of granule neuron soma from control (blue bars) or Pten mutant (red bars) was measured from sections stained with cresyl violet. CCI-779 treatment caused a significant reduction in the size of Pten-deficient neurons compared with vehicle treatment (P < 0.05).
