(A) Phenotype of OVA-specific memory CD8⁺ T cells at d 60 after priming with anti-DEC:OVA+ anti-CD40. OVA-specific cells were enriched using MACS sorting as described in the methods and gated as tet⁺ CD8⁺ cells. Naïve cells are not tetramer binding cells and were gated as CD44^low CD8⁺ cells. (B) Memory CD8⁺ T cells were identified in the indicated tissues: lung, intestine (IEL, LPL), Peyer's patches (PP), mesenteric lymph nodes (MLN), and spleen at d 60 after priming with anti-DEC:OVA+anti-CD40. Numbers are % of lymphocytes.
