Sensitivity of prion detection by amyloid seeding for preformed amyloid fibers or PTA-purified prions. (A) Preformed amyloid fibers were added to recMoPrP(89–230), and the mean lag phase for amyloid formation was measured. (B) Mean ThT fluorescence is shown as a function of the preformed seed concentration at 3 h. (C) Sc237 was added to recSHaPrP(90–231) in 10-fold serial dilutions, and the ThT fluorescence was measured at 19 h relative to controls. Total PrP in the Sc237 PTA pellet was calculated by ELISA; the 10−8 dilution contained ≈0.03 fg of PrP.
