Cell survival following alkylating and oxidative chemical challenge. (A) Vaco8 and SW948, but not Vaco411, possess intermediate resistance to MNNG. The three MSS mutators and two control cell lines (HCT116, MSI mutator and SW480, MSS nonmutator) were treated with 0–5 μM MNNG for 45 min at 37°C. The cells were grown for 6 days, subcultured and counted using trypan blue exclusion with a haemocytometer. (B) Vaco411 and SW948, but not Vaco8, possess resistance to H2O2. The same cell lines were treated with 0–300 μM H2O2 for 60 min at 37°C. The cells were grown for 5 days, subcultured and counted using trypan blue exclusion with a haemocytometer. Experiments were performed three times, each in triplicate and the error bars represent 1 s.e.m.
