ERK inhibition decreases bacterial phagocytosis by alveolar macrophages. A half million alveolar macrophages/ml were seeded unto chamber slides and allowed to adhere for two hours. The cells were exposed to U0126 (20 uM) or nothing for two hours and then opsonized GFP-e.coli. (25/cells) was added to each well. Following a 30 minute incubation the slides were washed X6 with PBS and adherent or phagocytosed bacteria were evaluated using fluorescent photomicrography or bacteria per cell was counted (50 cells per group).
