At the cell surface, MHC protein accumulates in distinct
microdomains. 221/Cw6-GFP cells were stained with
(A) phycoerythrin-labeled TÜ39 mAb (red),
(B) TÜ39 mAb followed by Alexa 568 goat anti-mouse
IgG mAb (red), (C) TR-transferrin (red), or
(D) biotinylated cholera toxin with
phycoerythrin-labeled streptavidin (red). No staining was seen with
phycoerythrin-labeled streptavidin alone. The green fluorescence of
HLA-Cw6-FP, each red fluorescence, and an overlay of both is shown.
(Right) Yellow indicates colocalization of green and red
staining. Magnification: ×6,000.
