Effects of androgens on the nuclear content of mature SREBP-1. After incubation for 2 days in the absence (−) or in the presence (+) of 10⁻⁸ M R1881, LNCaP cells were harvested and a crude nuclear fraction was prepared. Equal amounts of proteins from the nuclear fractions of duplicate dishes were subjected to SDS/PAGE and blotted onto a fortified nitrocellulose membrane. The blot was incubated with mAb IgG-2A4, directed against SREBP-1. Immunoreactive bands were visualized by using an ELC Western blot analysis system and a peroxidase-coupled goat anti-mouse secondary antibody. Positions of molecular size markers are as indicated.
