NGF induces BDNF release from PC12 cells. (A) Time course of BDNF release from undifferentiated PC12 cells before, during, and after application of 50 mM KCl or 100 ng/ml NGF. PC12 cells (3 × 105) infected with an adenovirus gene transfer system expressing BDNF were perfused as described in Fig. 2. BDNF contents, expressed in pg/ml, was measured with a two-site ELISA system. (B) Release experiments were conducted under conditions without constant perfusion, “static condition” by using 2 × 106/1.8 cm2 confluent PC12 cells. Cells were infected with about 5 × 106 plaque-forming units of an adenovirus gene transfer system expressing BDNF. After washing, the release of BDNF was initiated either by 50 mM KCl or 100 ng/ml neurotrophins (NGF, NT-4/5 or, NT-3) and BDNF content was determined by ELISA. Standard errors are referred to four independent experiments.
