HSV-1 Specific CD8+ Tcells are preferentially recruited to sites of viral replication. In H-2kB restricted C57BL/6 mice HSV-1 specific CD8+ T cells are almost entirely specific for the HSV-1 glycoprotein B immunodominant epitope SSIEFARL (100,101). Mice were infected with HSV-1 McKrae via corneal inoculation. At day 7 post-infection, leukocytes were taken from the (A) draining lymph nodes, (B) whole blood, or (C) the trigeminal ganglia. Cells were purified and analyzed by flow cytometry for expression CD8, CD45, and staining with phycoerythrin (PE) labeled H-2kB tetramer containing SSIEFARL peptide.
