3.L2 T cells from 3.L2tg×A72tg mice are still sensitive to antagonism after thymic and peripheral exposure to endogenous A72. Donor 3.L2 transgenic T cells from 3L2×A72 splenocytes were depleted of endogenous antigen presenting cells by passage over nylon wool columns and transferred to A72tg⁺ or A72tg⁻ recipient mice at a dose of 2.5 × 10⁶ CD4⁺CAB⁺ cells per mouse. Mice were immunized with 2 nmol Hb(64–76) peptide emulsified in CFA, and draining lymph nodes analyzed 5 days subsequently as described in Fig. 1. Results are representative of two experiments containing four total pairs of mice.
