(A) The NP34 transgene reduces the percentage of CD69+ thymocytes. Cells were isolated from F5+Rag-1−/−Tap-1+NP34+, F5+Rag-1−/−Tap-1−/−NP34+, positively selecting F5+Rag-1−/−Tap-1+, nonselecting F5+Rag-1−/−Tap-1−/−, and negatively selecting F5+Rag-1−/−Tap-1+I-E+ thymuses. Cells were stained with anti-CD69 phycoerythrin antibodies and analyzed by FACS. Bars represent the mean percentage of CD69+ thymocytes from three to eight mice of each genotype. SDs are indicated. (B) The NP34 transgene has no influence on the absolute numbers of thymocytes. Cells from F5+Rag-1−/−Tap-1+NP34+, F5+Rag-1−/−Tap-1−/−NP34+, positively selecting F5+Rag-1−/−Tap-1+, nonselecting F5+Rag-1−/−Tap-1−/−, and negatively selecting F5+Rag-1−/−Tap-1+I-E+ thymuses were isolated and counted.
