Effects of rosiglitazone (Ros) and 15d-PGJ2 (15d) on Ang II-induced TGF-β1/Smad2/3 and JNK signalling pathways in cardiac fibroblasts. Cells were pretreated with 5 μM rosiglitazone or 15d-PGJ2 for 1 h, and then stimulated with 0.1 μM Ang II for 24 h. Western blot analysis was subsequently used for detecting the expression of TGF-β1, Smad2/3 and JNK protein; cells were pretreated with 5 μM rosiglitazone or 15d-PGJ2 for 30 min, followed by 0.1 μM Ang II incubation for 20 min. Phosphorylated proteins were then detected. Rosiglitazone and 15d-PGJ2 inhibited Ang II-induced TGF-β1 expression (a), Smad2/3 (b) and JNK (c) expression and phosphorylation. All values are showed as mean±s.e.mean of three experiments. *P<0.05 vs control; P<0.05 vs Ang II.
