Effect of FVa on peptidyl substrate cleavage by membrane bound rFXaI16L. The initial rate of chromogenic substrate cleavage by membrane-bound FXaI16L (10 nm; 50 μm PCPS) was determined using increasing concentrations of SpecXa in the presence of different fixed concentrations of FVa (0 nm (▴), 1 nm (▵), 2 nm (♦), 5 nm (⋄), 8 nm (•), 12 nm (○), 18 nm (▪), and 20 nm (□)) in assay buffer. The lines are drawn following global analysis of the initial velocity data as described under “Data Analysis.” The data are representative of two similar experiments.
