NKT cells constitutively express IL-23R and RORγt mRNA, but expression of IL-17A requires triggering by receptor engagement. A, Naive T cells (Tn), memory T cells (Tmem), NK cells, and NKT cells were isolated from untreated mice by FACS. Constitutive levels of IL-23R and RORγt mRNA were quantitated in unstimulated cells using real-time PCR. Data were normalized to 18S and expressed relative to expression in naive T cells B, Changes in RORγt mRNA were measured 16 h after stimulation. Data are normalized to 18S and expressed relative to expression in unstimulated memory T cells. C, Changes in IL-17A expression in cell populations isolated from WT and IL-6 KO mice as measured by ELISA. All data represent at least three experiments that were highly reproducible. αCD3, anti-CD3.
