Effect of Cdk5 and p35 overexpression in pho85Δ cells. (A) Growth arrest caused by coexpression of Cdk5 and p35. pho85Δ strains transformed with pCDK5 or pCDK5-I with either a plasmid expressing p35 (p35) or an empty vector (230v) were grown in SD medium, serially diluted, and plated on glucose (SD-URA-TRP, Right) or galactose (SG-URA-TRP, Left) medium. (B) Morphology of pho85Δ cells overexpressing Cdk5 and p35. Cells were grown in raffinose-containing medium (noninducing) to an OD600 of ≈0.1, and then galactose was added to a final concentration of 2% to induce expression of CDK5 and p35.
