Complementation of JFH1 replicon by chimeric HCV helper viruses. (A) Five micrograms of the reporter RNA was transfected alone or cotransfected with 5 μg of the indicated helper virus RNA into Huh7-Lunet cells. Replication of the reporter replicon was assessed by luciferase assays of cell lysates harvested at the given time points. Mean values of duplicate measurements are given. (B) Release of infectious HCVTCP was quantified by inoculation of Huh7.5 cells with cell-free culture fluid harvested from transfected cells at 24, 48, 72, and 96 h posttransfection.
