TNF-α stimulation leads to a sustained decrease in Suv39h1 recruitment in db/db VSMC but not db/+. Suv39h1 occupancy at promoters of IL-6 (A), MCSF (B), MCP-1 (C), and MCP-1 enhancer (D) analyzed by ChIP assays using SUV39H1 antibody in MVSMC stimulated with 10 ng/ml TNF-α for indicated time points. Results were expressed as the percentage of untreated control (mean ± SE; *, P < 0.05; **, P < 0.01; #, P < 0.001; ***, P < 0.0001 vs. TNF-α-treated db/+, n = 3).
