Trypanosome cells were electroporated with FAT RNA or poly(I)⋅poly(C) as a control, and 1 hr or 16 hr after transfection, were pulsed for 30 min with 35S-radiolabeled amino acids. Radiolabeled proteins were separated by two-dimensional gels, and the positions of α- and β-tubulin were determined by using Western blot analysis with specific mAbs. The levels of radioactivity associated with α- and β-tubulin were determined by PhosphorImager analysis and are expressed as percentages of α- and β-tubulin synthesized in control cells, which are set at 100.
