Effect of Cd2+ on the level of ROS production in HEI-OC1 cells. FL1-H, green fluorescence. (A) Fluorescence measured by flow cytometry using DiOC6 in cells exposed to Cd2+ (20 μM) for 2, 8, or 12 hr; the right shift of cell distribution to the high fluorescence area indicates an increase of ROS production. (B) ROS levels (mean ± SE) in cells were also measured using the DCFH-DA fluorescent probe and a spectrofluorometer; the relative fluorescence levels were measured and plotted as a function of Cd2+ exposure time.
