Splenocytes from mice immunized with HEL plus IL-6 emulsified in IFA respond to subdominant and cryptic determinants of HEL, and dendritic cells isolated from lymph nodes draining the immunization sites (of HEL plus IL-6 in IFA) present the cryptic determinant to specific T cell hybridomas. Splenocytes from CBA (A; one of two experiments) and BALB/c (B; one of three experiments) mice immunized with either HEL in IFA (closed symbols) or HEL plus IL-6 in IFA (open symbols) were cultured with the proteins and peptides shown, and proliferation was measured as for Fig. 2. (C–E) B cells (circles) or dendritic cells (triangles) from BALB/c mouse lymph nodes draining immunization sites of either HEL in IFA (C), HEL plus IL-6 in IFA (D) or, from a separate experiment, ovalbumin plus IL-6 in IFA (E) were incubated at graded doses with cryptic-determinant (HEL 2–16) specific hybridoma cells at 5 × 104 cells per well with (closed symbols) or without (open symbols) HEL 2–16 peptide at 100 μg/ml for 24 h.
