Effect of extracellular variation of [Ca2+]free on Rho* phosphorylation in retina with closed α-toxin pores. After incubation of punches of retina with α-toxin in the presence of 1 μM [Ca2+]free and addition of [γ-32P]ATP in the dark, pores were closed by the addition of ZnSO4 and punches were transferred to a new incubation buffer containing variable [Ca2+]free (1 μM, 1 mM, or 10−10 M). The phosphorylation reactions were started at t = 0 by a flash.
