The Kd values were obtained by EMSA from the averaged data of four sets of titration gels. The number of active enzyme molecules in our preparation was determined by titrating the enzyme with the G/βFU-oligo under stochiometric conditions (oligonucleotide concentration = 10 nM > Kd). The equilibrium lies entirely on the side of the protein–DNA complex under these conditions, and the number of active enzyme molecules can be directly titrated.
