Regression parameters vs. library size. Regression parameters for the effects of nine variant amino acids were calculated for subsets of the libraries to determine how they varied as a function of library size. Five of the parameters covering the range of behaviors are shown. The subsets contained equal numbers of variants from the three libraries until a subset size of 360, at which point the 120 variants from the proline library were exhausted. The variants in the subsets were chosen in the order they were picked from the transformation plates.
