Stretching of DNA held with one side in an optical trap using continuous flow. (A) Schematic of the assay. DNA is attached to a bead with one end, the bead is held in an optical trap. To overcome the entropic forces that keep the DNA compact a continuous solvent flow is applied extending the DNA. DNA or associated proteins can be visualized using fluorescence microscopy. (B) Application of this assay to the formation of RecA filaments (40). λ-DNA is incubated with fluorescent RecA and filament formation is monitored by fluorescence.
