Amantadine isochronic inhibition curves for WT A/M2, D44A and S31N ion channels. Nine different concentrations of amantadine were applied to oocytes expressing WT, D44A, and S31N ion channels for 2 min at pH 5.5 and the current was measured before and after drug addition. Three oocytes were used for each drug concentration. The results were fit into the dose-response curve in GraphPad Prism 5. The best fit IC50 values for WT A/M2, D44A, and S31N were 16.0 ± 1.1, 15.8 ± 1.1, and 199.9 ± 1.1 μM, respectively.
