We performed alkaline methanolysis on scrapie brain spiked MBM as described under Methods and analyzed the reaction products as well as the inoculation samples by Western blot using monoclonal (mAb) 3F4. Results show no PrP detected in any of the 3 phases produced by the reaction (crude), nor in the inoculum we subsequently prepared by diluting each phase to 25 µg of total brain equivalent protein into 50 µl of 320 mM sucrose. After proteinase K digestion PrPsc was clearly detected in MBM spiked with infected brain but not subjected to the reaction (MBM sc, crude +PK). 1% NBH served as a control, displaying a slightly higher molecular weight, since it was not proteinase K digested.
