Samples and sorting strategy for TCR repertoire analysis. (b) Gating strategy used to sort KP9-specific CD8 T cells. Thawed pigtail macaque PBMC samples were stained with CD3, CD8, KP9 tetramer, CD45RA, CD28, and ViVid viability dye. Viable single CD3+ tetramer+ lymphocytes, and singlets were sorted into RNAlater with a modified BD FACSAria under biosafety level 3 conditions and used for TCR repertoire analysis.
