Biosynthesis of HFE proteins in COS-7 cells. After 60 h from the start of transfection, cells were pulse-labeled with ³⁵S-translabel for 0.5–4 h. At each time point, equal amounts of trichloroacetic acid-precipitable counts from cell homogenate were subjected to immunoprecipitation using CT16 IgG. The immunoprecipitates for wild type (○), C282Y (▵), and H63D (□) were analyzed by SDS/PAGE followed by fluorography. The fluorogram was quantitated by cutting and counting the gel pieces corresponding to the HFE bands. The quantitative results are shown (Lower).
