In vitro replication activity of the TrfA protein. Reactions were carried out under standard replication conditions as described in Materials and Methods with the use of the five-iteron plasmid pTJS42 and increasing amounts of wild-type His6-TrfA or the His6-TrfA 254D/267L protein as indicated. Total nucleotide incorporation was determined after 30 min. incubation at 32°C. (A) Replication with a crude extract (FII) prepared from E. coli C600.
