HSV-mediated silencing in primary neurons. Rat DRG neuronal cultures were transduced with a GFP-expressing vector (MOI = 1.0) and either HSV-U6shGFP or HSV-U6shLacZ (MOI = 5.0). Fluorescent microscopy at 48hrs post-transduction revealed that transduction with HSV-U6shLacZ had no effect on GFP expression levels, whereas transduction with HSV-U6shGFP resulted in a dramatic reduction in the levels of GFP compared to control groups (magnification 10×).
