The k_obs values represent the average of at least two independent trials, which differed by <30%. Reactions were conducted under single-turnover conditions in 100 mM KCl, 400 mM NaCl, 7.5 mM MgCl₂, 7.5 mM MnCl₂, 50 mM HEPES pH 7.0 at 23°C. The underlined region of S22 is identical to the bipartite consensus sequence, with exceptions denoted in boldface. The sequence of the substrate (sub.) is also provided, written 3′ to 5′ in opposite orientation. YrH represents the dinucleotide cleavage junction, where Y = C or T, and H = A, C or U.
