TatA cannot direct the degradation of NrfC mutants in the absence of TatBC. TatA was expressed in ΔtatABCDE cells using the pEXT22 plasmid, together with NrfC and mutants M1 and M2. The NrfC forms were induced by addition of arabinose as shown in previous figures, and samples were analysed by immunoblotting at times (in hours) after induction. The lower part of the figure shows a TatA immunoblot of a sample from one of these strains (the TatA/NrfC-expressing cells) and ΔtatABCDE cells expressing NrfC but not TatA.
