The effect on NF-κB activation of tmTNF-α-mediated reverse signaling by transfection of a mutant tmTNF-α plasmid or by treatment with sTNFR2. (A) Raji cells (1×10⁶) were transfected with pcDNA3.0 containing Δcs-tmTNF-α cDNA or the control empty vector. After a 48-h transfection, cells were harvested and lysed in lysis buffer. NF-κB activity was detected by ELISA. (B) Raji cells (1.5×10⁶) were incubated with different concentrations of sTNFR2, as indicated, for 30 min.
