Cre protein expression is dose and time dependent. C57BL6 mice were injected with specified titers of MUP-iCre-AAV8 or vehicle. At specific points, their livers were homogenized and analyzed for expression levels of Cre recombinase (38 kDa) by Western blotting. β-Actin (37 kDa) is the loading control. A: Cre protein is detectable 1 wk after injection of 1.4 × 1010 DRP/mouse MUP-iCre-AAV8 and increases over 1 mo. There is no Cre protein detected in vehicle or empty MUP-AAV8-injected mice. B: there are serial increases in Cre protein levels in mice injected with increasing DRP of MUP-iCre-AAV8.
