The tail domain of Myo2p is sufficient for localization. MYO2DN cells were grown in galactose for 0 (I), 1 (A–F), or 3 h (G and H), fixed, and stained using anti-HA antibody. The tail localized to the bud tip after a 1-h shift to galactose; 23 ± 7% of the cells demonstrated polarized Myo2p staining. After 3 h of growth on galactose, polarized staining was no longer observed. Scale bar, 5 μm.
