Solubilization characteristics of sperm membrane HK1-sc. (A) A membrane fraction (P100) from cauda epididymal mouse sperm was incubated in different extraction conditions as follows: 1) buffer (untreated); 2) 0.5 M NaCl; 3) 100 mM Na2CO3 at pH 11; 4) 2 mM glucose-6-phosphate; or 5) 1% Triton X-100. Only Triton X-100 resulted in the solubilization of HK1-sc. B) Triton X-114 phase separation of membrane-bound HK1-sc. A membrane fraction (P100) prepared from cauda epididymal mouse sperm was subjected to Triton X-114 phase separation (Bordier, 1981). HK1-sc immunoreactivity partitioned into the amphiphilic detergent phase and not the hydrophilic aqueous phase.
