Treatment with farnesyl transferase inhibitors improves growth and reduces nuclear blebs in fibroblasts expressing increased levels of wild-type lamin A. Fibroblasts expressing flag-lamin A, flag-progerin and GFP (control) at passage 13 were incubated in media containing the 10 nM farnesyl transferase inhibitor (FTI) L-744832 or in media containing the vehicle only (DMSO) for 14 days. Media was changed every four days. Cells were then analyzed for growth (A) and nuclear morphology (B) as described in Materials and Methods. Data shown represent mean +/− standard deviation of three independent measurements. Statistical significance between treatment groups was determined by a two-tailed student’s t-test. Asterisks: the data does not reach statistical significance.
