Blockade of IFNAR1 increases immature DCs in the PLNs of NOD mice. NOD mice were treated i.p. with two injections of anti-IFNAR1 or control mAb at days 15 and 20 after birth (n = 4 per group). On day 25, single cell suspensions were prepared from the PLNs, ILNs, and spleens, and stained with mAbs against CD11c, mPDCA-1, CD80, CD86, and MHC II and analyzed by FACS. (B) Splenic CD11c+ cells were magnetically enriched and stimulated with LPS (1 μg/ml) for 48 h in vitro. IL-10 or TNF-α levels in the supernatant were measured using an ELISA.
